The Basque Institute for Agricultural Research and Development, Neiker-Tecnalia, has developed a new method for the early detection of the diseases mildew, powdery mildew and botrytis in vines. The new methodology based on molecular biology techniques makes it possible to detect the disease before the symptoms appear on the plant. That way it is possible to carry out the rapid treatment of the plots or areas affected and prevent the disease from spreading all over vineyard, which reduces infective pressure. The R&D centre has also studied the evolution of infection by the pathogen Plasmopara viticola –the cause of mildew– on four vine varieties: Solaris, Cabernet Sauvignon, Tempranillo and Petit Courbu. Solaris turned out to have the greatest resistance to the fungus.

The early detection of diseases is fundamental for the agricultural sector. In the sphere of vine growing there are three diseases caused by fungi and which greatly affect vineyards: mildew, powdery mildew of the grapevine and botrytis, also known as grey mould or grey rot and which also affect other fruit crops. The fungi that cause them are Plasmopara viticola, Erysiphe necator and Botrytis cinerea, respectively. The three reduce crop yields and fruit quality, which leads to significant production and economic losses for the grape producing sector. This is compounded by the hazards arising out of the intense applications of plant protection products to combat these diseases, because they contaminate the soil and water in the vineyards and also pose a risk for the people who carry out the fumigation.

To spot the above-mentioned three diseases rapidly, Neiker-Tecnalia has fine-tuned a molecular biology technique based on Polymerase Chain Reaction or PCR; it is a method capable of detecting the genetic material of the relevant spores to confirm their presence or otherwise in the vineyard. This technique constitutes an alternative to the method based on identifying, under the microscope, the spores gathered in the vineyard by means of devices to capture the air, either volumetric or passive ones; in any case, this can only work a few days in advance of the appearance of symptoms on the plant, when the disease can by spotted with the naked eye. The problem facing this methodology is the contamination of the spore samples by remains of fungicides, dust from the ground, pollen and other matter. These elements are potential inhibitors of the reaction of the polymerase and therefore prevent the spores from being identified.

The PCR technique proposed by Neiker-Tecnalia includes the washing of the spores and ensures that they reach the DNA obtaining process in a clean state. Once the possible inhibitors have been eliminated, the R&D centre's researchers subject the samples to the PCR technique to identify the presence of the spores of Plasmopara viticola, Erysiphe necator and Botrytis cinerea and that way confirm each disease in the vineyard.

Solaris, the variety that best resists mildew

The other line of research conducted by Neiker-Tecnalia has been channelled towards studying the degree of resistance of mildew –a disease caused by the fungus Plasmopara viticola– which arises in the Solaris, Cabernet Sauvignon, Tempranillo and Petit Courbu grape varieties. The Solaris variety, of German origin, has turned out to be the most resistant, since it has a gene making it resistant to this fungus and is the one that takes longer to develop the infective process of the disease. Cabernet Sauvignon –a very widespread variety internationally– displayed intermediate resistance, and the least resistant to the pathogen were Tempranillo –the most common grape in the Rioja Qualified Appellation of Origin– and Petit Corbou –common in the south of France and in the vineyards of Txakoli (Basque white wine)-.

Metodologia berria biologia molekularreko tekniketan oinarritua da, eta aukera ematen du landarean sintomak agertzen hasi baino lehen detektatzeko gaixotasuna. Hartara, tratamendu bizkorra eman dakieke gaitzak erasandako sail edo eremuei, eta gaitza mahasti osora zabaltzea eragotzi; horren ondorioz, gutxitu egiten da kutsatze-presioa. Bestalde, ikerketa-zentroak aztertu du nola eboluzionatzen duen Plasmopara viticola patogenoak —mildiu gaitza eragiten duenak— lau mahats-barietatetan: Solaris, Cabernet Sauvignon, Tempranillo eta Petit Courbu. Onddoaren kontrako erresistentziarik handiena Solaris dela ondorioztatu dute.

Patologien detekzio goiztiarra funtsezkoa gertatzen da nekazaritza-sektorean. Mahastizaintzaren arloan, onddoek eragindako hiru gaitz dira mahastien etsai nagusiak: Mildiua, mahatsaren oidioa eta botritisa dira; azken horri lizun edo ustel grisa ere esaten zaio, eta beste fruta-arbola batzuei ere erasaten die. Gaitz horiek sortzen dituzten onddoak hauek dira, hurrenez hurren: Plasmopara viticola, Erysiphe necator eta Botrytis cinerea. Hirurek ere gutxitu egiten dute uzta eta fruituaren kalitatea, eta horrek ekoizpena asko murriztea eta galera handia izatea ekartzen du mahastizaintzan. Horrez gain, gaitzei aurre egiteko erabiltzen diren fitosanitarioen aplikazio bortitzetik eratorritako arriskuak ere badira, kutsatu egiten baitituzte mahastietako lurra eta ura, fumigatzen aritzen diren pertsonei arriskua ekartzeaz gain.

Aipatutako hiru gaixotasunak goiz detektatzeko, Neiker-Teknaliak biologia molekularreko teknika bat garatu du, PCR edo polimerasagatiko kateko erreakzioa deritzonean oinarritua; metodo hori gai da intereseko esporen material genetikoa identifikatzeko, eta, hala, mahastian baden edo ez jakiteko. Solaris, mildiuaren aurkako erresistentziarik handiena duen barietatea

Neiker-Tecnalia desarrolla un nuevo método para la detección temprana de mildiu, oidio y botritis en el viñedo

El Instituto vasco de Investigación y Desarrollo Agrario, Neiker-Tecnalia, ha desarrollado un nuevo método de detección temprana de las enfermedades mildiu, oidio y botritis, habituales en las vides. La nueva metodología, basada en técnicas de biología molecular, permite detectar la enfermedad antes de que aparezcan los síntomas en la planta. De este modo se puede llevar a cabo el tratamiento rápido de las parcelas o áreas afectadas y evitar así que la patología se propague en el conjunto del viñedo, con lo que se logra disminuir la presión infectiva. Por otra parte, el centro tecnológico ha estudiado la evolución de la infección del patógeno Plasmopara viticola –causante del mildiu- sobre cuatro variedades de vid: Solaris, Cabernet Sauvignon, Tempranillo y Petit Courbu. La especie que presenta una mayor resistencia al hongo resultó ser Solaris.

La detección temprana de las patologías resulta fundamental para el sector agrícola. En el ámbito de la viticultura existen tres enfermedades causadas por hongos que afectan enormemente a los viñedos. Se trata del mildiu, oidio de la vid y la botritis, también conocida como moho gris o podredumbre gris y que afecta también a otros cultivos frutícolas. Los hongos que las provocan son, respectivamente, Plasmopara viticola, Erysiphe necator y Botrytis cinerea. Las tres disminuyen el rendimiento del cultivo y la calidad de la fruta, lo que supone grandes pérdidas de producción y económicas para el sector vitícola. A ello hay que sumar los riesgos derivados de las intensas aplicaciones de fitosanitarios para combatir estas enfermedades, ya que provocan la contaminación del suelo y agua de los viñedos, además de suponer un riesgo para las personas que llevan a cabo la fumigación.

Para la detección rápida de las tres enfermedades mencionadas, Neiker-Tecnalia ha puesto a punto una técnica de biología molecular basada en la Reacción en Cadena de la Polimerasa o PCR, método capaz de identificar el material genético de las esporas de interés, para confirmar o no su presencia en el viñedo. Esta técnica supone una alternativa al método basado en la identificación al microscopio de las esporas recogidas en el viñedo mediante capturadores de aire, bien volumétricos o pasivos, y que en cualquier caso se adelanta hasta varios días a la aparición de síntomas en planta, momento en que el ojo humano es capaz de detectar la patología. El problema al que se enfrenta esta metodología es la contaminación de las muestras de esporas con restos de fungicidas, polvo de suelo, polen y otras materias. Estos elementos son potenciales inhibidores de la reacción de la polimerasa y por tanto impedir la identificación de las esporas.

La técnica de PCR propuesta por Neiker-Tecnalia incluye el lavado de las esporas y que estas lleguen limpias al proceso de obtención de ADN. Una vez eliminados los posibles inhibidores, los investigadores del centro tecnológico someten las muestras a la técnica de PCR para identificar la presencia de esporas de Plasmopara viticola, Erysiphe necator y Botrytis cinerea, y de esta manera constatar cada enfermedad en el viñedo.

Solaris, la variedad más resistente al mildiu

La otra línea de investigación de Neiker-Tecnalia se ha dirigido a estudiar el grado de resistencia al mildiu –enfermedad causada por el hongo Plasmopara viticola- que presentan las variedades de uva Solaris, Cabernet Sauvignon, Tempranillo y Petit Courbu. La variedad Solaris, de origen alemán, ha resultado ser la más resistente, ya que cuenta con un gen de resistencia a este hongo y es la que desarrolla de una forma más lenta el proceso infectivo de la enfermedad. Cabernet Sauvignon –variedad muy extendida internacionalmente- presentó una resistencia intermedia y las menos resistentes al patógeno fueron Tempranillo –uva con presencia mayoritaria en la Denominación de Origen Calificada Rioja- y Petit Corbou –habitual del sur de Francia y de los viñedos de Txakoli-.
